Optimum Sterilization Method for In Vitro Cultivation of Dimorphic Seeds of the Succulent Halophyte Suaeda aralocaspica

Suaeda aralocaspica is an annual halophyte in the Amaranthaceae saline deserts of central Asia. This plant has succulent leaves and grape-like fruits a potential horticultural plant. To obtain efficient sterilization method optimal culture conditions, two types seeds produced from single S. were treated with 75% ethanol for different time durations first, then sodium hypochlorite (NaClO) or mercury chloride (HgCl2), five timing treatments used second seed surface sterilization. Sterilized germinated on Murashige Skoog (MS) medium at hydrogenation (pH) levels, to examine germination seedling performance. The results showed that highest percentage brown was 100% black 17%. Thus, more suitable further experiments than seeds. For seeds, effect NaClO better HgCl2, based germination, contamination, survival. Rinsing 60 s, sterilizing 8 min, cultivating pH 8.0 MS 7 days best all procedures cultivation methods tested, which been successfully applied vitro culture. optimized protocol described here can be as reference genus.
